Hello,
I have been using the 3D OC macro to find the sum of fluorescent intensities of stacks, specifically series of different nuclei stained with DAPI. I have been having a problem where the macro ignores objects of a certain size. I have tried adjusting the size filter to much higher than the region of interest should be, and it still does not work. It picks up everything but the large areas, and I have had it work on ROIs 100 times the size of the objects it excludes, so I can’t figure out what is going on here. Any advice would be much appreciated!
