Take an image with your lens showing an object with a feature of a defined size. I use the grid of a hemocytometer for this. Take images of your object with any available lens and keep them for future reference.
If you have the image of your object, use the line tool to draw a line of defined length, then use Analyze, Set Scale... "Global" to define the length of the line you have just drawn. After this you can draw Scale bars on this and other images using Analyze, Tools, Scale Bar...

Subject: Adjusting microscope scale plugin
Dear Forum,
I want to put microscope scale into my picture since my microscope could not be using to put scale bar. I read about microscope scale plugin of image J, however I don't know how I can customize my microscope setting with this plug in. My microscope is Olympus BX-50 with ocular lens is 10x and objective lens are 4x,10x,20x,40x,and 100x. How I can adjust the setting of this plugin with my microscope condition, since I dont know much about java program, any easy solution would be very appreciated.
