Hi Saar,
In the Threshold window, *don't* click Apply. Then the original pixel
values will be retained.
You also have to check "Limit to Threshold" in Analyze > Set
Measurements.
Another way to do it, where you don't have to keep the threshold
window open, is to set the threshold values, then Edit > Selection >
Create Selection and all the pixels within the threshold will become
an ROI.
To measure thresholded pixels within another ROI, add your other ROI
and the "create selection" ROI to the ROI Manager. Then select both
and More >> AND them.
