Hi,
I am about to quantify the amyloid deposition formation using area
fraction. I am given a number of slides already scanned at high resolution
and I am about to use the cavalieri point-counting method. I'd like to
understand if the vast number of plugins and macros of ImageJ already
support what I need. The simplified workflow for one image is this:
1. (Manually) define a ROI (e.g. polygon or freehand selection)
2. Systematically overlay a small number of rectangular counting regions
with a rectangular (crosshair) grid in them.
3. For each counting region in turn, vertices found to be within the
amyloid plaque are manually marked.
4. Marked vertices are appropriately counted, area fraction is
determined.
Any pointers would be appreciated to existing plugins and macros so that I
don't have to re-implement things from scratch. Ideally I would like that
- With the counting frame density and grid density defined, once the ROI
is specified the counting frames are autogenerated and placed on the image
as overlay(s)
- Marking vertices is supported by mouse-click (obviously), aided by
snap-to-grid (large cursor area), and possibly paint mode (with cursor
pressed, all vertices are marked which the cursor sweeps).
- Mark and unmark (undo) is supported.
