Best option to segment cells in dense cultures

Hello,

which is the best method to identify individual cells in dense
cultures? I am trying to count perinuclear/ cytoplasmic dots per cell in
dense cultures.

Surya,

Any chance you can attach an image showing what you're looking at and
what you're trying to count and assign to particular cells?     I can't
tell from your description, for example, whether the cells are somewhat
translucent and you are looking at a culture more than one cell thick
and trying to match the dots in 3D, not just 2D.
