Can I select based on Pixel Intensity? Nuclear Vs Cytoplasmic Quantification

Dear ImageJears,

I am trying since few days, without success, to have answers to two
questions regarding ImageJ:

1) Can I select based on Pixel intensity ? (I mean SELECT ALL PIXELS
FROM THE IMAGE which are BLACK or WHITE or HAVING SPECIFIC INTENSITY (or
a RANGE) ?)

2)Where can I change the threshold (or any other parametesrs) for Magic
Wand?


I want to measure (quantify) Nuclear Vs Cytoplasm  ratio intensity in
immunofluorescence stained cultured cells.

This is what I do:

A ) For Nuclear Part:
I take the Hoechst (DNA only) slice and threshold it - and this gives
sharp good edges - then if I use Magic Wand I can select the nuclei.
Copy and Paste it to the Slice which I want to measure the area and
intensity (the SLICE whith the protein of interest)

If I hit UNDO, ONLY the SELECTION REMAINS into the NEW SLIDE. This is
excellent because now I have traced Nuclei and I can press M to measure


B) For the CYTOPLASM PART (THIS IS WHERE I NEED HELP)
If I do the same procedure as above, because the edges of the cells are
not so sharp, the MAGIC WAND DOES NOT WORK IN SELECTING PROPERLY (as I
have to keep SHIFT and SELECT)
I would like to SELECT ALL THE WHITE or BLACK PIXELS (or a SPECIFIC
RANGE ) and HAVE THE
SELECTION use in the real image in which I want to measure the whole
cell area and mean intensity.

Alternatively we can use VNC so you can see my screen and see what I am
talking about.
