Counting cells passing through a specific region

Hi Everyone,

I'm a new user of Imagej. I've spent days in the counting of cells passing through a specific region. I have stack and video taken from a light microscope. It is easy to cout cells in a single image. I'm wondering how to count cells passing through in the video or a stack. Is ther a plugin or method to realize this purpose? Any help and advice is greatly appreciated.
