Counting fliuorescene nuclei

Dear,

I have an experiment with NLS::GFP construct. The positive result is the fluorescence nuclei with black backgroud. I would like to quantify them using ImageJ but I don't know how to do it. I will really appreciate if you have solutions for me. I attach one file for an example

PS: I am a mac user

I am not sure my way works for you or it is correct, but that what I do in my lab to score IF images:
   1. Go to PROCESS> BINARY or IMAGE>TYPE>8BIT
   2. Go to IMAGE>ADJSUT>THRESHOLD to set up lower and upper threshold
   3. Use ANALYZE > MEZSURE to know the pixels for different shapes of AIM to count
   4. Go to ANALYZE > ANALYZE PARTICLES and the window
