Counting particles within a droplet

Hi All - I'm fairly new to ImageJ and wondered if any of you experts could help me solve a problem. I have taken some images of droplets containing fluorescent particles and would ideally like to be able to get:

a) a droplet count by size class
b) a particle count per droplet within that size class

Am I hoping for a bit too much?

It struck me that it is not too dissimilar to measuring granules within a cell, so hopefully someone might be able to offer some advice to a newbie!
