Counting two types of cells

Hello, I am searching for a way to automatically count two types of differentially stained sperm cells.
One type is red, the other green, the pics are taken under a fluorescence microscope.

Without examples, it is hard to tell whether just "Image>Color>Split
Channels" followed by a threshold segmentation and particle counting would
be good enough.
