DNA Quantification

Cordy
Hi, I was wondering if anyone could help me. I am trying to quantify ng of
DNA using my ladder as standard. Should i be using the area under the plot
from the gel analysis menu or the profile plots?

Any help you could provide would be greatly appreciated.

Best wishes,

Cordy

Hi Cordy,

I think the gel analysis menu (strg 1/2/3) is the right choice.

As you probably want a standard curve / "line", it might be necessary to convert your image to some sort of pseudo OD, from ij1.38o, there is a nice function in the calibrate menu - thanks Wayne!.

As you have your standards in the scan, actually every method should do it, as long as you treat your standards and the samples the same.

Best regards,

Wolfgang

Thanks Wolfgang!

I guess you're right any method should work as long as I treat my
standards the same. I will give several methods a try and see which one
gives me the most reproducible data.

Best wishes,

Cordy
