Detecting cell spheroid on image with analyze particles

Hello,

I wish to detect a cell spheroid in a picture taken by standard light microscopy. Despite having a proper threshold which locates the spheroid, the command "analyze particles" always detects objects from the opposite colour. For example, if I apply a threshold which "stains" the spheroid white, the command "analyze particles" detects dark regions. Now if the threshold "stains" the spheroid in black, the same option will detect objects in white. Is there a way to reverse this effect by modifying the "analyze particles" command?

*The picture is an RGB image which is converted to binary after thresholding.

Hello Mikael,

I believe that if you're converting to binary after thresholding, color or otherwise, Analyze Particles chooses as "countable" the areas that have a value of 255, regardless of color.  

You can tell which value your spheroid has in the binary image by holding the cursor over it and reading out the value from the bottom line of the taskbar.  If your spheroid has the 0 value and your background has 255, try Edit --> Invert and see if you get the desired result.    

I've searched for documentation on this and can't find it, but I'm sure someone else will correct me if I'm wrong and / or point out the correct documentation.
