Help: setting a scale - not in the simple way!

Rotem Oved
hello everybody,i'm trying to measure length using Fiji. However, the images created by the microscope do not have a scale bar in them. Is there a way I can set the scale properly, converting pixels into nm?I tried using "Bio-Formats Importer" to open the image, but still, the length shown in "all paths" window doesn't match the length that shows up after performing the "analyze skeleton" (I added a photo for example).I'm sure I can retrieve the information because it does appear (hopefully, the correct data) at the "all paths" window.thank you so much.

Hi,
I think that you can set the pixel dimension by image->properties here you can set the unit and the equivalent pixel to your unit of length.

To convert from pixel to your unit of length, you can use this formula
PIXELonTheImage = pixel_of_the_microscope*(binningFactor/TotalMagnification))

Hoping to be helpful,
