How can i make measurements for confocal images ?

I have images in .tif format.
These images were obtained using Zeiss confocal laser scanning microscopy.
I have to make certain measurements in micrometers.
Does anyone know how to do this ?

The images should retain their calibration from the microscope.  If they have retained their calibration then you will see at the top of the image their measurement such as "112.50x112.50 μm". Then you select the straight line tool and drag it across what you need to measure.  Then go to Analyze -> Measure and the value in the Length column is what you're looking for.
