How to automatically select all cells in image without thresholding

Hi.
I know there are plugins and such out there that can select all cells with thresholding, but how can I do it automatically without thresholding? I have images with many small cells and I want to basically select the cells from the original image and find the average greyscale value within each cell, so I can't have it thresholded otherwise the greyscale value is changed. You can use a freehand selector to draw around a cell manually and then use the measure function. I want to be able to do that for all cells automatically. Any help or specific macro/plugin suggestions would be appreciated. Thanks!
