How to split a/two freehand roi into four quadrants?

Hello everyone.
I am thinking about doing cell immunofluorescence polarization analysis.

does anyone know how to automatically split a freehand roi at the its centroid. and splite into four quadrants becoming four rois that i can quantify the fluorescence intensity? (left)
i was even thinking that there could be two rois, one the cell, one the nucleus. and the centroid for splitting could be based on either the centroid of the cell or the centroid of the nucleus.


but i've been searching the userguide and it said only one selection possible.
and i have tried composite selection by overlaying a freehand roi and two lines and combine in roi manager and failed...

Could anyone kindly point me to the right direction how i automate this?

I have the exact question with almost similar goals.

Essentially, how do I divide an ROI (could be a drawn oval, in its simplest form) into four quadrants (axes in the shape of an 'x') so I can measure pixel intensity in each of the quadrants?

Did you ever find a solution?

It would be great if you or anyone can share. Thanks!
