Image Analysis using Image J

Hello,
       I am a new user of the Image J software and had few queries on the
application I am looking for. My issue is as stated below:

"I have Calibration pictures that were taken at 40X lense and with different
zooms on the camera say 50, 77, 88 etc. These images taken need to be
calibrated from pixels to microns in the Image J software if possible. Then,
the stained images of the tissue taken with the same lens and zoom under the
same miscroscope need to be added in the software and the thickness and
diameter need to be measured in microns."

Can this be done with Scion imaging software? Could you please advice me on the
same? If its not possible can anyone please suggest me an alternative?

relatively straight forward if you do it manually.  You would just use the
calibration images - drawing straight segmented lines across your known
calibration grid or sphere distance - and the "Analyze -> Set Scale" menu to
calculate the calibration numbers for each zoom setting.  Then when you open
an image at a certain zoom and magnification, you would just go to that same
menu item and enter the calibration number for those settings. Then you would
just draw straight segments across your tissue structure and use the "Analyze
-> Measure" to get your measurement.
