ImageJ: Measure RGB areas

Hi All

I have histological slides that I would like to measure the RGB  
areas. Is there an easy way to do it without splitting the RGB? (I  
have tried RGB measure plus plugin, but it gives RGB means, not the  
colour areas).

Hi Richard,

I assume that you have more than one colour that you want to measure the
area of.

I don't know which histology stains you are working with but most of
them seem to be difficult to separate out just based on RGB. You do need
to do segmentation if looking at more than one colour. Splitting the RGB
channels would be the easiest way to do it in my opinion if that
distinguishes the stains sufficiently well.

Otherwise, I would suggest trying the Threshold Colour plugin (available
at Gabriel Landini's site
http://www.dentistry.bham.ac.uk/landinig/software/software.html). I find
that HSB is really useful for separating histology stains based on hue.

An alternative approach is the Colour Deconvolution plugin
(http://www.dentistry.bham.ac.uk/landinig/software/cdeconv/cdeconv.html
), since there are a number of stain combinations already available
there. It worked brilliantly for me for H&E and DAB/Haematoxylin or
DAB/Nissl without having to do any adjustments.

Once you have the separate images, it's straightforward to go ahead and
measure your areas.
