Hi,
My name is Martin Ward and I am a researcher from the university of
Edinburgh. I have been attempting to use image J and the Trackmate
plugin to track particles with the aim of extracting useful values
from their brownian motion (effective radius, diffusion etc.).
However I have been unable to track the particles at all. I have
followed the tutorial, and all works well with the test data. In
appearence, my input looks very similar to the test data (bright white
spots on a dark background), however when I get to the final stages no
tracks are found.
I have spent the past couple of days playing around with the settings
and levels of filtering etc, however this has not improved the
situation. I am not sure at all why this is happening, or indeed how
to rectify the issue. Any help at all on this will be greatfully
received. Also, if you require a sample of my input files, feel free
to let me know and I can send something to show.
Also, I remain unclear on what the final output from Trackmate is?
Will it, in the end, spit out the sort of values that I am looking for
(Diffusivity, radius of gyration etc.) or must the plotted tracks then
be further analysed to extract these values later?
