I have a tiff image, and I created an ROI and then saved the selection. Later I reopened the original tiff image, and then reopened the saved ROI, and a yellow outline of the ROI was added to the image. When I adjust threshold (without applying to segment) and then Analyze particles (with "Limit to threshold" checked in the Set Measurements), sometimes the analyzed particles are limited to the ROI and sometimes it ignores the ROI and counts particles throughout the image. I can't figure out what is different about the times it works and the times it ignores the ROI.
Any ideas?
