Measurement of DNA with DAPI

Manuel Bonilla
I am trying to use Image J for measuring the amount of DNA in an algal cell. How can this be done?

Have a look at this paper:

David C. Hardie, T. Ryan Gregory, and Paul D.N. Hebert, “From Pixels to
Picograms: A Beginners' Guide to Genome Quantification by Feulgen Image
Analysis Densitometry,” J. Histochem. Cytochem. 50, no. 6 (June 1, 2002):
735-750.

If I recall correctly the paper has a curious comment suggesting not doing
background correction. I am not sure that it is right, but the rest of the
paper is very informative.
