Measuring Skeletonized Objects vs. Non-Skeletonized Objects

Hi All

I am attempting to obtain measurements of transmittance and diattenuation
from images collected using polarization microscopy. The objects that we
are measuring exhibit different properties on their edges when compared to
their centers. We have tried performing a binary skeletonization on the
objects, but do not feel that that correctly represents what we are
attempting to measure. Does anyone have any experience with measuring the
properties of an object with and without skeletonization?
