Hi all,
So, what I am trying to do is to measure the size, in microm3, of cells, in Image J.
I acquire the images with a confocal microscope:
resolution : 512x512
pixel size : 28.8 microm
step size : 150 microm.
What I do is this :
- Split the colors, in order to isolate the blue signal (DAPI)
- Image>Stack>Reslice : I set the output spacing to 5.2 (150/28.8 = 5.2)
- I then use the 3D object counter plug in. It gives me a size of 4 470 294 pixels^3.
I multiply that by 0.0288^3 and get 107 microm3.
What do you think? Is this legit, or am I doing something wrong somewhere?
