Measuring sea Ice

Hello,
I need to measure the area of sea ice from satellite picture . First I have to select the area in witch I have interest with polygon and to cut the background from all images .After I have to make histogram for all images to count how many white pixels I have in the area. I have a folder where are my images. They are png. I tryed some macros or to run Batch Process but I have no success . I have the coordinates of the zone of interest in txt. file, when i marked the Polygon I save the coordinates in txt. . I forger to mention that all pictures are of the same area of the globe for each day from 2006 to now and from them I need to know the number of white pixels in one zone .
