Need advice on active profiling

Dear List,

I want to measure the degradation rate of an object in a time lapse series and have been advised that an active profiler would be the way to do this.  I am using the ABSnake plugin and have successfully segmented an object in that it is outlined in red accurately as it shrinks in size.  However, when trying to measure its size change using  ROI Manager the area does not change over time (but the integrated density does).  After saving the stack when I open it the red-outlined segmented ROI is not recognized as a selection by ROI Manager, making it difficult to do further analysis.  

Any advice on how to achieve my goal of quantifying the degradation rate or correctly using ABSnake is welcome.
