Hello ImageJ Community,
I have a question regarding the Plot Profile tool and how it generates
values for fluorescence intensity. When a line selection of more than 1
pixel in width is drawn through a confocal image containing something
fluorescent, ImageJ should create a dataset where the "x" values represent
the length of the line and the "y" values represent the average intensity
of the pixels. This works very nicely for a line that is drawn
horizontally. If that same line is drawn vertically or diagonally,
however, the values that end up in the "y" column are not a simple average
of the pixels at any given position on that line. So in summary, I am not
sure how ImageJ calculates those values for lines that are not perfectly
horizontal. Any assistance would be greatly appreciated. Thank you very
