Quantification of RAW images (Nikon NEF)

Dear list members,

 

I would like to quantify brightness of structures from images taken in RAW
format (Nikon NEF, no compression). I am not sure about data conversion /
processing regarding quantification / linearity issues.

 

Is there any ImageJ plugin (or other standard software) to convert the RAW
image to standard TIFF and /or to extract individual color channels from the
raw file directly without sacrificing linearity?

 

Is this issue covered in detail somewhere on the Web?

it gives several options how a RAW image can be converted.
