After selecting the first ROI, hit the 't' key to add it to the ROI
Manager.
Then select the second ROI, call Edit>Selection>Make Inverse to select
exactly the opposite (i.e. everything that was not selected), again add it
to the ROI Manager with 't'.
Then select the both added ROIs in the ROI Manager, click on More>> and
select "AND" (which is a computer programmer's way to say "intersection").
That should add the ROI you desire to the manager as a 3rd ROI which you
can activate by clicking on it. You can even click ROI Manager's Measure
button to measure what's inside the ROI.
