Ratiometric analysis of Confocal Live cell Images

Hi,
     I'm beginner in the world of Image J and I want to analyze the confocal Images of Live cells which express a protein that is excited at two different wavelengths but the emission wavelength is the same. I want to do a ratiometric analysis of the images acquired at the two different excitation wavelengths.
                 
Can anyone please let me know the step by step procedure to either obtain the results graphically or any other way format?
