Re: Scale Bar LSM format image

Sindy Kueh-2
Dear Image J users

I would like to add scale bar to images captured using the ZEN 2009 software. Images are store as LSM files. Unfortunately, the output scale seemed to be incorrect.
I have attached a picture of muscle fibres, showing the scale bar on bottom left which is incorrect: each fibre diameter is estimated to be in the range of 20-40uM, Do I need to calibrate Fiji - pixel to uM?

Thank you
Sindy

Re: Scale Bar LSM format image

Cammer, Michael
In our experience with ImageJ, LOCI plugins or Bioformats version, and Zen 2009, as long as the lens was set correctly in the Zen software, the spatial scales and time intervals import fine.
Perhaps you could post one of the native lsm files?

Re: Scale Bar LSM format image

Sindy Kueh-2
Hi Michael
I tried to attached a file, but it is too large even when compressed. Interestingly, when looking for a suitably sized file.... I noticed that the issue was not apparent with my smaller sized files. The scale bar was correct.
Is there any way I could upload the problem images to you or image J list?

Thank you
