Relative movement of objects and contour extraction

Hello everybody,

 

I'am new to the field of ImageJ and thus I'am not so familiar with all the
great plugins that are established. I have fluorescent images of a spherical
"soap bubble"  that gets more and more buckled by a small particle (on the
right from the center of the image).

 

Now I have two concerns:

 

1.)    The position of the right boundary of the spherical object changes
relative to the position of the small particle which is fixed. The green
arrow in the images indicates the position of the right boundary of the
"bubble". Comparing all three pictures, the boundary shifts to the right
relative to the center of the fixed particle. How can I extract this shift
for a whole image stack? Is there an appropriate toolbox existing? I tried
to track the maximum of a line profile running across the right boundary of
the bubble without hitting pixels from the particle. But then my result
depends on the position where I calculate the profile.

 

2.)    Is there a way to extract the contour of the ellipsoidal/spherical
"bubble" including the curved area around the particle? I'am interested in
extracting the curvature of this object and the cross-section of the bubble.
