Selecting Cells: transfected GFP-tagged, untransfected, background

Dear List,

I would like to be able to take a plated layer of cells and automatically
distinguish three types of cells/areas in the image:

-transfected cells (gfp signal),
-untransfected cells (either non-specific dye or just outlines), and
-background (no cells)

Is there currently a plugin or otherwise rigorous approach to doing this? I
have tried to think of some clever ways to do this, but it seems like it
must be such a universal problem that someone must have already addressed
and solved it. One could just do this by pixel, but that might get dicey
when cells move/warp/change slightly over time.
