Simple method of measuring total fluorescence

I am looking for a simple method of comparing protein absorption on a surface (boat coating) compared to a reference surface. Basically total FL-intensity in rectangular box of x times y microns. The protein is FITC marked. Any input would be appreciated.

If you a have an image, you can define multiple ROIs and measure them simultaneously. You will get a table with the intensity which ou can analyze in any other program.
Just make sure that you select "integrated density" in "set measurements".
