Hi,
1. Start ImageJ
2. Mark and drop the two images into the ImageJ window (from your local explorer/finder)
3. Select from the Menu "Process -> Image Calculator…"
There you will find "Subtract" and … Possibly the result should be 32-bit, allowing also negative result values!
Hopefully that is a solution for the problem not only for the subtraction!
