Thresholding an ROI without duplicating the image

Hi,
If I have an image with about 400 ROIs and I would like to threshold each
one of them separately, how would I go about doing it? I read forum posts
that suggested duplicating the image each time I'd like to threshold an
ROI. Since I have about 400 ROIs per image and many images, I need a
different solution.

What can I do instead?
