I use TrackMate to segment spots of different intensities and sizes and i want measure the total intensity of these spots.
I use the Log detector and then define the estimated blob diameter and the threshold. I did not modify any other parameter. Spots are detected, diameters seem to be correctly measured (small spots have small diameters and big spots big diameters) but the result of the total intensity measures makes no sense: total intensity is the sum of all the pixels inside the blob and the blob size is invariant for all spots, disregarding whether they are big or small.
Someone knows how can I get the total intensity of the real spot inside the blob and not of all the pixels inside the blob?
I know Trackmate is recognizing the sptos inside the blob because the estimated diameter in the results is lower than the diameter of the blob and accord to the size of the spot.
