Hello, I am trying to figure out the best way to count each particle in a stack. I tried attaching the stack, but the file was too large (if anyone would like, I could send it through my email). Basically it is accumulations of discrete particles that accumulate in a furrow. This particles aggregate, and I need a way to figure out how many go to the first, second and third cleavage furrows. I have been using "Analyze particles" and checking by eye counts. Counts are very high (anywhere for 1K to 3K), and I do not necessarily agree with the counts that I am getting from analyze particles.
This is what I have been doing: stack to z-project, then threshold, analyze particles, find a few particles (singles and clear doubles, triples...etc.) and average them finding the "size" of one, finally I use that average to figure out how many particles in the image
Is there a better way to count the particles? I have been playing around with the 3D objects counter, but I am not sure I am doing it in the right way.
