calculating the surface of a 3D object in an Image J stack

Dear ImageJ users I am a scientist currently working in the UK and also an
NIH Image turned to ImageJ user , and I would like to ask if any of you
knows how to calculate surfaces of 3D objects from ImageJ stacks .  For
example , imagine you have a stack of z sections of a round cell whose
outer membrane has been brightly fluorescently decorated and whose inside
has been dimly decorated . In 3D this is like a sphere which is bright on
the surface and dim inside . How could one isolate the surface in order to
calculate specifically the fluorescence of the surface of sphere ?  Thank
