Dear ImageJ list,
We are some new users of ImageJ trying to compare the color density of
individual cells which have been stained, fixed on slides, pictured using
microscopic camera (each picture contains approximately 30 cells). We have
tried to convert the pictures from color to 8-bits, eliminating the
background by thresholding, free-hand tracing cell boundaries to measure
the integrated density of each cell. However we encounter the following
problems and would like to know if anyone in the server list could give us
some advice.
1. The results of the integrated density are not consistent. For
example, 2 cells which are visually different in color density would have
closed integrated density values. Is there any plug-ins out there that
could compensate this problem? Or is there any feature in the original
ImageJ that will do?
2. The result of the mean & integrated density would change sometimes
when it is measured twice (without alternating anything). Anyone out there
who encountered the same problem before? And how did you solve it?
3. Is there any automatic cell-tracing feature in ImageJ?
