I need to determine the percent of new bone matrix in association with DBM. I can come close by masking all else but ( all the) bone and then, because new bone has nuclei and DBM does not, I can count nuclei - at least theoretically. This almost works but there are too many other similar specs ( H&E ) . However, those specs are usually separated by 100s of microns or are stringy, while new bone nuclei are typically within 20-50 micron of each other. The strings can be filtered using Particle analysis.
So, the real questions :
Is there an algorithm / macro that can select items ( nuclei) within a certain distance of each other ?
Once this is done, is there a way to select and add a specific area around each nuclei ? e.g. add touching areas within a 50 micron radius of the nuclei. This would give a good approximation of new bone. "excess" area outside the bone could be masked using the original image.
