I am very new to righting macros...
We are measuring auto radiograms of in situ (35s)
First I have set the calibration using Rodbard to optical density.
I have a Stouffers 21 step transmission density scale, which I have imaged with our scanner.
Measured the values 1-21 entered the actual values 0.06-3.0
To measure the background we multiply a user selection (background) value by 3.5 and add the
mean,to define the signal.
This is put in the results window. Ok
Now I want to get that number "BackGround" (something like 0.22 depending on the section)
and put it in to the Threshold menu 0-0.22.
This will create a mask to measure the signal under.
How do I get the "BackGround" value from the results window in to the threshold menu?
Then i would like to subtract the "BackGround" value from the rest of the measurements made on
that section. Then calculate the Integrated Optical Destiny with the "Background" corrected mean
as Corrected IOD.
When I record the process, I get 0-255 gray values in the macro record window.
