measurement of fluorescence intensity over time

María Victoria Pepe
Hi!

I need to measure the fluorescence intensity within a circular selection
and then, repeat the measurement in the same way over 800 slices (each one
represents a time point)

I would like to know how to create a macro that allows me to do this
automatically

Good day María Victoria,

I guess you need the mean intensity in the circular selections for all
slices.

If so,

1.
make the desired selection on the first slice of your stack

2.
add the selection to the ROI Manager
(Analyze >> Tools >> ROI Manager >> Add)

3.
makes sure that in "Set Measurements" the "Mean gray value" is checked

4.
in the ROI Manager dialog go to "More >> Multi Measure"

5.
in the opening dialog check all options and click "OK"

6.
a Results table opens showing the desired mean values

Hi María Victoria,

if you have an image stack, you can simply use Image>Stacks>Plot z-Axis
Profile. No macro required.
Alternatively, use Image>Stacks>Measure Stack; it will give you the
values for whatever properties you have selected in Analyze>Set
Measurements. You will probably want "Mean".

Of course, you can record these commands (Plugins>Macros>Record) and use
them in a macro.
