Hello Michaels +Kees,

Many thanks for getting back to me!

I have had a go with moving the hue sliders and I think that will definitely work- hurrah!  My new stumbling block is how I get a total area measure for all of the green areas combined (these now being black on a binary photo).  When I click Analyse> Analyse particles, a new window pops up asking me to enter values for size (0- infinity) and circularity.. so perhaps I should just be clicking Analyse>Measure instead?
Secondly, do I need to click Process>Find edges before trying to work out area??

Finally, Michael (Cammer), I am a bit of an Image J novice so I need a little more direction re your response- where do I need to paste your code into?

Dear Sarah,

I have attached a little macro that will report the file name and the percentage of black pixels in your image. Save it in the directory macros and run it via Plugins-->Macros-->run (or Plugins-->macros-->install and the macro will appear for this session in the Plugins-->macros menu for faster access). This can easily be further developed into a macro to analyse all binary images in a directory or as part of a larger analysis. Hope it does the job.
