question about autobrightness settings?

Hi,

My name is Micah, a PhD student and having an issue with quantifying GFP
image in image J. When I open this file (attached), the brightness and
contrast differ between what you see in preview and Image J. I believe
the preview appearance is correct because thats what I saw under the
microscope also. Has anyone run into this issue? Some how there is an
auto brightness setting that is changing the way my image appears in
ImageJ.

Help appreciated!!

Hi Micah,

Short answer:
* Image > Adjust > Brightness/Contrast
* Set
* Min -> 0, Max -> 65535
* Now it will match what you see in Preview

Alternate approach:
* File > Import > Bio-Formats
* Choose your file
* Uncheck the "Autoscale" box
* Click OK
* Now it will match what you see in Preview
