Best fit polynominal / get coordinates from ROI line

Hi
I have some OCT images and would like to perform regression to 2 Roi lines (curved) to a best fit polynominal in order to calculate de mean distance between them and the area.
coul anyone help me since I can not get the coordinates nor execute a best fit to a line ( I use de Wand tool)
