Using Image J to count live and dead cells in cultures

Hi all

I was wondering if amnybody could help me with a query regrding cell counting using image. I've stained my cultures for dead and live cells and now want to quatify them. I have tried using several methods on image J including adjusting the threshold, watershed and also downloaded a pluggin (ITCN) but for some reason i'm still not able to count the labelled cells accurately.

Any help or suggestions in regards to this will be greatly appreciated.
