Thanks for the suggestions. I will try them.
However, I suspect thresholding and enhanced depth of field may not be sufficient to overcome the
UNEVEN surface of the tissue, which results in target nuclei from the topmost epithelial cell layer and
undesirable nuclei from lower levels in the same z-slice. All are in focus and have similar size and
intensity. Perhaps I can differentiate them with shape-based thresholding. Any ideas?
