With the Lab-stack frontmost select "Type > 8 bit" from the "Image"-menu.

Then, from the same menu, select "Color > Stack to RGB"

Finally make your ROI-selection in the resulting Pseudo-RGB image and
select "Tools > Color Histogram" from the "Analyze"-menu.
