Dear All,
I have to calculate the calcium conc. using the Grienkiwicz
equation. I am not able to understand the difference between the calculation
of Beta and Rmin, Rmax, as from the reports I have read I can see both are
ratios but then is it that Beta is calculated only on slide without the
sample while Rmin and Rmax are deciphered from slides loaded with sample. I
am confused in this regard . Someone please help.
[Ca ]i = KD x ß x (R -Rmin) / (Rmax- R)
I have no idea of the difference between
ß = fluorescence signal at 351nm at 0
μmol.l-1 Ca2+ / fluorescence signal at 364 nm at 39.8 μmol.l-1 Ca2+.
Rmin = ratio at 0 mmol.l-1 Ca2+.
Rmax = ratio at 39.8 μmol.l-1 Ca2+.
