measure the "shortest path" ?

Dear all,
I´m trying to measure the distance of immunogold-labelled proteins in
cell using electron microscopy images. The experimental pertubation
should affect the distance of the particles from the cell membrane.
questions:
1.) is there something like a "leat-cost-routing " that automatically
(or semi-automatically) finds the shortest path e.g. from the particle
to the membrane ?
2.) could the membrane be "marked" as the reference to which the
shortest path should be generated (the membrane would be represented by
a line then) ?
